| To the best of our knowledge, little information is available regarding the presence of Anaplasma species in camels in Iran. This study sought to investigate the presence of Anaplasma species by microscopy and polymerase chain reaction (PCR) assays in 100 healthy dromedaries (Camelus dromedarius) arriving for slaughter. The microscopic examination of Giemsa-stained blood films revealed that Anaplasma like structures could be identified in the erythrocytes of two blood smears. To confirm the presence of and to identify the species of Anaplasma spp., a PCR technique was performed using primers amplifying a 750 bp fragment of the 16S rRNA gene of Anaplasma and the PCR products were analyzed by sequencing. The nucleotide sequence was compared to the sequences available in GenBank using the Basic Local Alignment Search Tool (BLAST). According to the results, the sequences of two 16S rRNA PCR products clearly fit within the Anaplasma genus in the family Anaplas mataceae. In this study, phylogenetic analysis using the 16S rRNA gene sequences revealed that two sequences obtained from monophyletic clusters included Anaplasma ovis (A. ovis). The obtained sequences had 99.6-100% similarity with previously published 16S rRNA gene sequences. This study aimed to evaluate the presence of novel genetic variants associated to A. ovis in dromedaries in the world. | اطلاعات اندکی در خصوص وجود گونههای آناپلاسما در شترهای ایران در دسترس است. در این مطالعه یک صد شتر یک کوهانه به ظاهر سالم که برای کشتار به کشتارگاه فرستاده شده بودند برای شناسایی گونههای آناپلاسما بوسیله آزمایش میکروسکوپی و آزمایش واکنش زنجیرهای پلیمراز مورد بررسی قرار گرفتند. آزمایش میکروسکوپی اسمیرهای خون محیطی رنگ آمیزی شده با گیمسا نشان داد که دو نمونه خون از یک صد گسترش خونی دارای ساختارهای آناپلاسما مانند بودند. جهت تایید وجود و شناسایی گونههای مختلف آناپلاسما، آزمون واکنش زنجیرهای پلیمراز با استفاده از آغازگرهایی که یک قطعه 750 جفت باز از ژن 16S rRNA را تکثیر میکردند انجام و محصول آزمون تعیین توالی شد. توالی نوکلئوتیدی بدست آمده با استفاده از نرم افزار بلاست با توالیهای موجود در بانک ژنی مقایسه شد. تجزیه و تحلیل دو توالی بدست آمده از ژن 16S rRNA نشان داد که محصولات مورد نظر در جنس آناپلاسما و خانواده آناپلاسما تاسهآ قرار دارند. تجزیه و تحلیل فیلوژنتیک با استفاده از توالی ژن 16S rRNA نشان داد که توالیهای به دست آمده در این مطالعه در خوشه واحدی با آناپلاسما اوویس قرار دارند. نتایج حاکی از شباهت 100-6/99 درصدی توالیهای تکثیر شده با توالیهای منتشر شده ژن 16S rRNA آناپلاسما اوویس می باشد. مطالعه حاضر حضور واریتههای ژنتیکی جدید مرتبط با آناپلاسما اوویس را در شتر یک کوهانه در جهان گزارش کرده است. تحقیقات تکمیلی برای یافتن ناقلین و همچنین اهمیت دامپزشکی و پزشکی این واریته به ظاهر جدید در ایران نیاز می باشد. | ||
