چکیده
Fowlpox (FP) is a viral disease that is widely distributed throughout the world. The disease has an economic impact on the poultry industry, and its prevalence has even been reported in vaccinated flocks. The present study used flow cytometry to evaluate the CD4+ and CD8+ T-cell immune response of chicks induced by FP vaccine. 120 specific pathogen-free (SPF) 21-day-old chicks were randomly divided into three groups of 40. One group was used as negative control with PBS inoculation, the other two groups were inoculated with the local fowlpox vaccine produced by Razi Institute and commercial FP vaccines, and they were kept for five weeks. Peripheral blood mononuclear cells (PBMC) were isolated using Ficoll–Hypaque density gradients and the percentages of CD3+, CD3+, CD4+, and CD3+CD8+ T lymphocytes were analyzed with flow cytometry. Seven days post-immunization, a maximum (90-100%) swelling formation (“take”) on the vaccination site was observed. The ratios of CD4+ to CD8+ T-lymphocytes in both vaccinated groups were significantly higher (p < 0.05) than the control group inoculated with PBS. The percentages of CD3+, CD3+CD4+, and CD3+CD8+ T-lymphocytes were increased in chickens vaccinated with commercial and local FP vaccines. There were no significant differences between the groups receiving commercial and local fowl pox vaccines. The present study showed that protective immunity could be associated with increased cellular immune responses, which has been interpreted as enhancing T-cell proliferation and increasing CD4+ to CD8+ ratios through vaccination with the FP vaccine. This study further suggests that the induction of enhanced immune responses is due mainly to the Th1-type response.

یابی پاسخ ایمنی سلولهای CD4 T- و T-CD8متعاقب تزریق واکسن آبله طیور در جوجه های SPF به روش فلوسایتومتری

آبله طیور یک بیماری ویروسی است که گستردگی جهانی دارد. بیماری آبله سبب خسارت اقتصادی بر صنعت طیور میگردد. شیوع آن در گله های واکسینه شده نیز گزارش شده است. هدف از این مطالعه بررسی پاسخ ایمنی سلولی جوجه های عاری از پاتوژن (ُSPF) پس از تزریق واکسن آبله طیور با روش فلوسیتومتری می باشد. تعداد 120 جوجه SPF (21 روزه) به سه گروه 40 تایی به صورت تصادفی تقسیم شدند. یک گروه به عنوان کنترل منفی با تلقیح PBS و دو گروه دیگر با واکسن آبله طیور ساخت موسسه رازی و واکسن تجاری تلقیح شدند. سلولهای تک هسته ای خون محیطی (PBMCs) از نمونه های خون هر گروه حاوی EDTA، با استفاده از گرادیانت فایکول جدا شدند. درصد جمعیت سلولی لنفوسیت های T CD3+، CD3+ CD4+ وCD3+ CD8+ با استفاده از روش فلوسایتومتری مورد تجزیه و تحلیل قرار گرفت. نتایج این مطالعه نشان دادکه حداکثر 7 روز پس از واکسیناسیون تقریبا تمامی نمونه های واکسینه شده دارای تورم در محل تزریق بودند. گروه های واکسینه شده نسبت به گروه کنترل بیان بالاتری از لنفوسیت های T را داشتند. درصد جمعیت سلولی لنفوسیت های CD4+ و CD8+ در جوجه های واکسینه شده با واکسن تجاری و واکسن موسسه رازی افزایش یافت و تفاوت معنا داری بین دو گروه واکسن تجاری و واکسن موسسه رازی وجود نداشت (P < 0.05). نسبت لنفوسیت های CD4+ به لنفوسیت های CD8+ در هر دو گروه از گروه های واکسینه شده، به وسیله واکسن آبله طیور موسسه رازی و واکسن تجاری، از گروه شاهد که تنها به وسیله PBS مایه کوبی شده بودند به طور قابل توجهی بالاتر بود. نتایج مطالعه حاضر نشان داد که پاسخ سیستم ایمنی جوجه ها پس از تلقیح واکسن آبله با افزایش فعالیت سیستم ایمنی سلولی همراه است و که به شکل تکثیر سلول های لنفوسیت T و افزایش نسبت CD4+ به CD8+ (به دلیل تزریق واکسن آبله طیور) را سبب شده است. در این مطالعه مشخص گردید که بطور عمده القاء افزایش واکنش های ایمنی توسط سلولهای نوع Th1 اتفاق میافتد.
