چکیده
Leptospirosis is a serious zoonotic infection and the most prevalence disease is in the tropical and subtropical region. The definitive diagnosis of Leptospirosis, caused by spirochetes of the genus Leptospira infection is already using culture methods, serological tests such as the microscopic agglutination test (MAT) and molecular detection methods (PCR) are possible.  In this study, we used multiplex PCR method for detection of pathogenic and non - pathogenic Leptospira based on lipL32 and 16S rRNA genes. All serovars were obtained from the Leptospira Reference Laboratory of Microbiology Department, Razi Vaccine and Serum Research Institute, Karaj, Iran. The PCR product for the lipL32 and 16S rRNA genes was 272 bp and 240 bp respectively. The sensitivity amplification for the multiplex assay was 10-6 pg / μl for 16S rRNA gene and 10-4 pg / μl for lipL32 gene. The sensitivity for multiplex PCR was 10-3 pg / μl. The results supported the idea that multiplex PCR can be used to detect Leptospira samples. This method was also able to differentiate between saprophytic and pathogenic leptospires and was able to do so much easily than conventional methodologies. Due to the slow growth of Leptospira and the importance of time in diagnosis, molecular methods such as PCR are suggested.

تشخیص مولکولی لپتوسپیراهای بیماری زا و غیر بیماری زا بر اساس ژن های lipL32 و 16S rRNA با استفاده از روش Multiplex PCR

لپتوسپیروز یک عفونت زئونوز است که توسط اسپیروکت بیماری زایی به نام لپتوسپیرا ایجاد میشود. جهت تشخیص زودهنگام لپتوسپیروز، تمایز گونه های بیماری زا و غیر بیماری زا از یکدیگر اهمیت ویژه ای دارد. توالی های ژن 16S rRNA برای شناسایی گونه های لپتوسپیرا مفید هستند. LipL32 یکی از مهمترین پروتئین های غشای خارجی لپتوسپیرا است که تنها در سرووارهای بیماری زا وجود دارد و فاکتور مناسبی جهت شناسایی لپتوسپیراهای بیماری زا می باشد. بنابراین هدف از انجام این پژوهش، تشخیص مولکولی لپتوسپیراهای بیماری زا و غیر بیماری زا بر اساس ژن های lipL32 و 16S rRNA با استفاده از روش Multiplex PCR می باشد.
سرووارهای بیماری زا و غیر بیماریزای لپتوسپیرا در محیط کشت اختصاصی EMJH کشت داده شدند و DNA ژنومیک آن ها تخلیص گردید. در این تحقیق از پرایمر 16S rRNA برای سرووارهای غیر بیماری زا و پرایمر lipL32 برای سرووارهای بیماری زا استفاده شد.
با توجه به داده های PCR مشخص شد که ژن 16S rRNA با سایز 240 bp فقط در سرووارهای غیر بیماری زای لپتوسپیرا حضور دارد و همچنین ژن lipL32 با سایز 272 bp نیز فقط در سرووارهای بیماری زای لپتوسپیرا وجود دارد و در سرووارهای غیر بیماری زای لپتوسپیرا مشاهده نگردید.
میزان حساسیت برای ژن 16S rRNA، 10-6 pg / μl و برای ژن lipL32، 10-4 pg / μl به دست آمد. میزان حساسیت نیز در روش Multiplex PCR 10-3 pg / μl بود.
نتایج نشان داد که یک آزمایش تشخیصی مولکولی مناسب با ویژگی و حساسیت بالا مانند Multiplex PCR و با استفاده از ژن های lipL32 و 16S rRNA جهت تشخیص و تمایز سرووارهای بیماری زا و غیر بیماری زا از یکدیگر بسیار کارآمد می باشد.
