چکیده
Clostridium novyi (C. novyi) causes deadly Black disease in sheep and rarely in other animals. Alpha toxin (α-toxin), the most apparent pathogen of this disease, is produced by C. novyi type B. Economic damages of C. novyi include sheep mortality costs, depreciation of affected farms, and health problems with infected carcasses. The identification of C. novyi and isolation of its pathogens by conventional methods is a time-consuming process, necessitating a simple and rapid method for isolating and detecting pathogenic C. novyi. Therefore, this study aimed to molecularly identify α-toxin in local C. novyi isolates from the sheep livers. In this study, 75 livers suspected of Black disease were sampled. The samples of the liver were cultured under anaerobic conditions. Some of the cultured colonies were used in biochemical tests. For molecular confirmation, the DNA of isolates was extracted, and the isolates were confirmed by the polymerase chain reaction (PCR) on the liver tissue and cultured samples using specific α-toxin primers. The PCR on α-toxin produced a band in the range of 609 bp, indicating that the samples belonged to C. novyi. According to the results, of 75 isolates, 18 isolates were confirmed as C. novyi. C. novyi type B was isolated from the liver and confirmed by biochemical and molecular characterization. The PCR assay ensured a sensitive and specific tool for the detection of C. novyi in the samples.

شناسایی ترکیبی مولکولی و بیوشیمیایی آلفاتوکسین کلستریدیوم نویی بومی جدا شده از کبد گوسفند

کلستریدیوم نوویی عامل بیماری کشنده هپاتیت عفونی نکروزان در گوسفند و ندرتاً در سایر حیوانات می‌باشد. بارزترین فاکتور بیماریزایی آن آلفا توکسین (α-توکسین) است که توسط نوع B تولید می‌شود. خسارات اقتصادی بیماری شامل هزینه‌های مرگ و میر گوسفندان، استهلاک مزارع درگیر و مشکلات بهداشتی لاشه‌های آلوده است. تشخیص بیماری‌های کلستریدیایی و جداسازی عامل بیماری با روش‌های مرسوم وقت‌گیر می‌باشد، لذا نیاز به یک روش ساده و سریع برای جداسازی و تشخیص کلستریدیاهای پاتوژنیک می‌باشد. بنابراین، هدف از مطالعه حاضر، شناسایی ملکولی توکسین آلفا در جدایه‌های بومی کلستریدیوم نوویی از کبد گوسفند است. در این مطالعه، از تعداد 75 نمونه‌ کبد، مشکوک به بیماری قانقاریای عفونی کبد جمع‌آوری شد. نمونه های کبد در شرایط بی هوازی کشت داده شدند. تعدادی از کلنی های کشت داده شده برای تست های بیوشیمیایی مورد استفاده قرار گرفتند. برای تأیید مولکولی،DNA جدایه‌ها استخراج شد و جدایه‌های بافت کبد و نمونه‌های کشت شده با واکنش زنجیره‌ای پلیمراز (PCR) و با استفاده از آغازگرهای اختصاصی α-توکسین، از نظر مولکولی بررسی شدند. با انجام PCR برای α-توکسین، باندی در محدوده 609 جفت باز ایجاد گردید و مشخص شد که نمونه‌های مورد بررسی به کلستریدیوم نوویی تعلق دارند. براساس نتایج، از 75 جدایه، 18 جدایه به عنوان کلستریدیوم نوویی تایید ملکولی شدند کلستریدیوم نوویی نوع B از کبد جدا شد و با خصوصیات بیوشیمیایی و مولکولی تایید شد. روش PCR ابزاری حساس و خاص است که تشخیص کلستریدیوم نوویی در نمونه‌ها را تضمین می‌کند.
