| To evaluate the effect of Metformin, Hypericum perforatum L., and Trigonella gracum seeds alone and combined with chitosan nanoparticles on streptozotocin-induced diabetes in rats (to compare the effects). In this study, 96 adult male Wistar rats were used, which were divided into 12 groups: Control, diabetic, diabetic mellitus receiving buffers, pure chitosan nanoparticles, Metformin at a dose of 250 mg/kg, hydroalcoholic extracts of Hypericum perforatum L. flower at a dose of 200 mg/kg and Trigonella gracum seed at a dose of 100 mg/kg alone and in combination, nano-extract of Hypericum perforatum L., nano-extract of Trigonella gracum seed, alone and in variety. The healthy group received flower extracts of Hypericum perforatum L. and Trigonella gracum seeds at 300 mg/kg. The biochemical test parameters of major liver enzymes, blood glucose¸ weight, malondialdehyde (MDA), plasma iron reducing capacity (FRAP) and antioxidant enzyme superoxide dismutase (SOD), and histopathological changes in liver tissue were determined. The diabetic groups treated with metformin and chitosan nanoparticles containing two extracts alone and in combination, significantly improved rat weight, liver enzyme index of alkaline phosphatase (ALP), MDA, and antioxidant enzyme SOD (P value ≤ 0.05). Chitosan nanoparticles containing the selected extracts combined affected glucose levels, showing more significant improvement than the diabetic groups treated with the extracts alone (P value ≤ 0.05). The livers of diabetic rats treated with an extract of Trigonella gracum seeds alone and with chitosan nanoparticles and with a combination of two parts of Trigonella gracum seeds and Hypericum perforatum L. at a dose of 300 mg/kg alone and in combination with chitosan nanoparticles showed significant improvement in histopathological changes compared with diabetic rats. It seems that the hydroalcoholic extracts of Hypericum perforatum L. and Trigonella gracum seeds, combined with chitosan nanoparticles, are good candidates for further evaluation as effective factors for future control of diabetes. | ||

| هدف از این مطالعه بررسی تأثیر دانه های متفورمین، Hypericum perforatum L. و Trigonella gracum به تنهایی و همراه با نانوذرات کیتوزان بر دیابت ناشی از استرپتوزوتوسین در موش صحرایی (به منظور مقایسه اثرات) بود. در این مطالعه از 96 موش صحرایی نر بالغ نژاد ویستار استفاده شد که به 12 گروه کنترل، دیابتی، دیابتی دریافت کننده بافرها، نانوذرات کیتوزان خالص، متفورمین با دوز 250 میلی گرم بر کیلوگرم، عصاره هیدروالکلی گل Hypericum perforatum L. با دوز mg/kg 200 و دانه Trigonella gracum با دوز 100 mg/kg به تنهایی و به صورت ترکیبی، نانو عصاره Hypericum perforatum L.، نانو عصاره دانه Trigonella gracum به تنهایی و به صورت ترکیبی تقسیم شدند.. گروه سالم دریافت کننده عصاره گل Hypericum perforatum L. و بذر Trigonella gracum را به میزان 300 میلی گرم بر کیلوگرم دریافت کردند. پارامترهای آزمایش بیوشیمیایی آنزیم های اصلی کبد، گلوکز خون، وزن ، مالون دی آلدئید MDA)، ظرفیت کاهش آهن پلاسما (FRAP) و آنزیم آنتی اکسیدانی سوپراکسید دیسموتاز (SOD) و تغییرات هیستوپاتولوژیک در بافت کبد سنجش شد. گروههای دیابتی تحت درمان با متفورمین و نانوذرات کیتوزان حاوی دو عصاره به تنهایی و در ترکیب، وزن موشها، شاخص آنزیم کبدی آلکالین فسفاتاز (ALP)، MDA و آنزیم آنتیاکسیدانی SOD را به طور قابلتوجهی بهبود بخشیدند دادند (P value ≤ 0.05). نانوذرات کیتوزان حاوی عصارههای انتخابی ترکیبی بر سطوح گلوکز تأثیر گذاشتند و نسبت به گروههای دیابتی که تنها با عصارهها تیمار شده بودند، بهبود قابلتوجهتری را نشان دادند (P value ≤ 0.05). کبد موش های دیابتی تیمار شده با عصاره دانه Trigonella gracum به تنهایی و با نانوذرات کیتوزان و با ترکیب دو قسمت از دانه Trigonella gracum و Hypericum perforatum L. با دوز 300 میلی گرم بر کیلوگرم به تنهایی و در ترکیب با نانوذرات کیتوزان بهبود قابل توجهی در تغییرات هیستوپاتولوژیک در مقایسه با موش های دیابتی نشان داد. به نظر میرسد عصارههای هیدروالکلی دانههای Hypericum perforatum L. و Trigonella gracum همراه با نانوذرات کیتوزان، کاندیدای مناسبی برای ارزیابی بیشتر بهعنوان عوامل مؤثر در کنترل آتی دیابت باشند. | ||
